'Function and development of dendritic cell subsets' - Lecture by prof. Kenneth Murphy

Prof. Kenneth Murphy (Washington University School of Medicine, St. Louis, US) will cover three areas central to effective anti-viral/tumor immunity

An update of recent findings on WDFY4 in cross-presentation by cDC1 (Theisen et al., 2018) will be given, followed by discussion of new work done since. I will discuss mechanisms of cDC1 licensing, briefly reviewing Ferris et al., (2020) and then presenting recent unpublished work. Lastly, I will discuss development of cDC1. After reviewing essential background, I present unpublished work based on several newly generated mouse mutants. Starting with our recent discovery of the Zeb2 -165kb enhancer (Huang et al., 2021), I will then present new mutations in this enhancer that show how Nfil3 represses Zeb2 to drive cDC1 specification. Inadvertently from this work, we accidentally generated a mouse with a complete loss of cDC2 progenitors and lack of peripheral cDC2s, representing a first genetic model complete cDC2 ablation. These mice fail to make TH2 responses when infected by H. Polygyrus, with massive egg burden, total lack of granuloma formation, and impaired TH2 responses.
