In our lab we design different synthetic polymers, i.e. macromolecules composed from a number of identical repeating units, to engineer the immune system. Innate immune activation is an important is an important hallmark in fighting against cancer and can be provoked by potent small molecule agonists of pathogen associated molecular patterns (PAMSs). However, uncontrolled PAMP triggering evokes systemic inflammatory immune responses and has hampered clinical translation of several classes of small molecule immune-modulators such as imidazoquinoline TLR 7/8 agonists. Taking advantage of the inherent albumin-binding property of lipid motifs and their tendency to accumulate in lymphoid tissue, we design amphiphilic lipid-polymer conjugates that confer suppression of systemic inflammation but provoke potent lymph node immune activation.
In our work we provide a rational base for the design of such lipid-polymer amphiphiles for optimized lymphoid targeting.
